Hey guys, so I was about to start the selection for my target this semester but realized that it's not a guarantee whether my target (KRAS) is biotinylated or not. This is obviously important because I want to know whether I should perform a bead or filter selection for this target. But this question probably applies to everyone's targets, to what extent should we trust the labeling on the freezer for biotinylated and non-biotinylated?

ReplyDeleteHave you emailed Melissa or Ashley D.? They both worked on the selection, so they should have a good idea of what's going on. Plus -they're both still around campus & should be pretty easy to get in touch with.

Yes I just talked to Ashley and she said there's a His tag that I could use nickel beads with. But she said it's possible to still do a filter selection due to KRAS's size and referred me to you on which type of selection I should perform. What do you think?

ReplyDeleteIt's your call! This is your project & you can take the lead. Think about it & try to figure out what would work best or if there is a better choice ... there may not be. My first thoughts are that either method would work.
